Lady of Slaughter is a side quest in Assassin’s Creed Origins in the town of Yamu.
Your task is to help Menehet find a guy (Pamu) who’s supposed to take part in the fight.

Summary of Lady of Slaughter
1. Find and bring Pamu to Menehet
2. Speak to the High Priest to replace Pamu
3. Defeat Isfet
Find and bring Pamu to Menehet
Your first objective is to look for Pamu.
You can find him in the market area of Yamu. As you get there, use Senu to pinpoint his exact location.
You should see Pamu at the village tavern. Carry him.
You can use your camel to transport him. Bring him all the way back to the Temple of Sekhmet to meet Menehet.
As you approach Menehet, a cutscene should load.
Speak to the High Priest
As Pamu is unfit to fight, you will replace him. But first, you need to inform the High Priest about the replacement.
He is located nearby, in the location shown below.
Talk to the man with the leopard costume.
Enter the ring to face Isfet’s fighter
Just walk forward at this point.
Defeat Isfet
Your main objective is to defeat Isfet at the battle arena. Hold “R2” to use the heavy attack to bypass his shield.
That will bring his shield down for awhile, and keep him on the ground a couple of seconds. While he’s vulnerable, keep attacking him.
You can also use the Overpower skill during the match to defeat him quickly.
The quest completes after you’ve defeated Isfet.
